Abstract
Molecular mechanisms involved in the stimulation of NGF secretion by cytokins (IL-1beta, IL-4, IL-6) and neurotransmitters (histamine) were studied on the model system of neonatal rat cortical astrocytes and cortical neurons in primary culture. In the astrocytes IL-1beta and histamine are potent stimulators of NGF secretion. Stimulation is mediated by IL-1beta and histamine H1-receptors, respectively. Cortical neurons do not respond to the stimulation by above substances.
We identified and characterised specific histamine H1- and H2-receptor binding sites on our model system of astrocytes and compared their molecular properties with the H1- and H2-receptors identified in vascular system. For their characterisation we used new H1- selective agonists (derivatives of 2-phenylhistamine and histaprodifen).
On the model system of isolated rat peritoneal mast cells two groups of drugs (tricyclic antidepresants and non-steroidal anti-inflammatory drugs (NSAIDs)) were studied for their ability to stimulate or inhibit histamine and serotonin release.
