Abstract
Changes in expression of potato plant defence-related proteins, particulary proteinase inhibitors, as well as certain other pathogenesis-related proteins will be studied at the transcriptional and/or translational level before and after infection with PVYNTN virus, as well as before and after attack. by Colorado potato beetle. As important plant defense-proteins, known and novel proteinase inhibitors will be isolated, characterized and cloned. After testing for their inhibitory activity on PVY virus protease and some insect digestive proteinases, cDNAs corresponding to selected inhibitors will be inserted into potato genom by Agrobacterium-mediated gene transfer and transgenic plants will be generated and tested on the resistance against PVYNTN virus and Colorado potato beetle.
