The injury of isolated rat heart induced by reperfusion was decreased by calcium antagonist mibefradil and by combination of vitamine E and aspirine. EqT II induced decrease of coronary flow was dependant on the presence of Mg++ and Ca++ and on Mg++/Ca++ ratio. By the methods of nonlinear dynamics we calculated the correlation dimension (D2) and maximal Lyapunov exponent what enabled us to suggest the classification of irregular rhythms of postischaemic and posthypoxic isolated hearts. In isolated coronary artery it was found that the effects of both enantiomers and racemic nicardipine did not depend on the presence of vascular endothelium. For the contraction of isolated porcine coronary artery the presence of L-type of calcium channels is necessary. Equinatoxin II dose-dependently and time-dependently caused histamine release from isolated rat peritoneal mast cells in vitro. The experiments, where the influence of antidepressants (AD) on histamine kinetics in the rat was studied, showed that AD can significantly diminish the levels of plasma histamine after its liberation from the stores in the body or after an administration of exogenous histamine. The kinetics of histamine elimination from plasma was also changed. We continue with the reasearch and we have begun also with the studies in the cat. The experiments on isolated portion of guinea pig stomach and on isolated guinea pig atrium showed that the choice of the type of isolated organ significantly influences the parameters of concentration-response relationship for histamine, very probably due to different experimental conditions. The experiments, performed on the isolated mouse trachea showed that this isolated preparation is a useful model for studying the effects on muscarinic or serotonergic receptors but it is less suitable for studying histamine receptors because of its low sensitivity to histamine.
