Abstract
Purpose of the project is to establish a full circle of a development of novel antimicrobial drugs - gyrase inhibitors. The project will integrate biotechnological techniques of cloning and isolation of recombinant bacterial gyrases, computational design of novel drugs based on the tertiary structure of the enzyme, organic synthesis of novel compounds, in vivo and in vitro testing of their inhibition of the gyrase and determination of tertiary structure of complexes by X-ray crystallography and NMR. Goal of the project is the synthesis of a novel gyrase inhibitor based on the rational drug design.
