Abstract
Summary of Objectives 1. Cloning of the genes coding for, and purification of, the Mur synthetases from pathogenic species (MRSA, staphylococci, enterococci, H. influenzae,…). 2. Kinetic analysis of inhibition of these enzymes 3. Determination of the 3D structure of selected enzymes and enzyme-ligand complexes. 4. Design, synthesis and bioactivity evaluation of potent inhibitors of these enzymes. 5. Preclinical profiling and toxicological evaluation of the best compounds.
