Significance for science
Inflammatory diseases, where TLR receptors are involved, and lipid oxidation are between main causes of death as they are involved in cardiovascular to autoimmune diseases. Identification of endogen ligands and molecular mechanism contributed to the development of the research area, which is prerequisite for development of efficient therapies and diagnostics. Cooperation with groups from Faculty of Medicine UL, University Medical Centre LJ and Medical university Vienna contributed to clinically important issues, where the results and clinical picture can have direct influence on patient´s treatment. Successful application of research of those diseases increased our knowledge on chronic diseases. Understanding of the molecular mechanisms is not only of medical, but also of biotechnological importance. Our group has a great expertise in the design of TLR receptor inhibitors, where we have already identified and patented compounds with a potential for treatment of inflammatory diseases (inhibitors of TLR4/MD-2 activation). Describing a role of TLR4 in sterile inflammation could broaden the application of these compounds and increase the possibilities to treat these disorders. Using innovative strategies and new ideas we educated students (undergraduate and PhD students) for research. In terms of education also our contribution to promotion of natural science can be included: In the period of last three years members of the project team have achieved exceptional successes in the competitions of research projects in the most eminent academic competition (iGEM, described in achievements). Thus, they importantly help to the promotion of natural science in the broad public and Slovenia as the state with the excellent science and education. A wide response of mentioned successes was encountered through all over the world in the newspapers, journals, TV, radio, on web pages.
Significance for the country
Despite advances in intensive medicine the mortality due to SIRS in sterile inflammation and chronic diseases increases. So, any advance in understanding of molecular mechanisms and drug design is therefore valuable. In the project we identified endogen ligands to describe molecular background of inflammation. Meantime, we already search for potential inhibitors of signaling and compounds for microvesicle neutralization. This opens a new pathway to combined therapy that would on one hand decrease MV presence in blood and on the other inhibit signaling of the remaining MVs. There is a long way from identification of leading compounds to new drugs, so there is a chance to find new collaborators or interested companies through european projects. Our results have from medical point of view noticeable impact on human health. Basic research usually leads to new discoveries, which then become basic for applicative research. So, the potential users of our research results, such as pharmaceutical industry, could use our basic results for further applicative research, as they open new horizons and subsequentially new views to problems. Within basic research new research methods will be introduced which could also serve as a know–how for industry to solve their problems.
