The effect of electroporation on the expression profile of genes in malignant melanoma cells is currently unknown. We demonstrated that the expression profile of tumour supressor genes or oncogenes is not affected by electric pulses used in electrochemotherapy or electrogenetherapy. Our results support the evidence that electroporation is a safe method and does not affect gene expression profile in melanoma cells.

Tumor vasculature is due to the specific proterties that makes is different from normal vasculature, an attractive target for treatment. In the study, we demonstrated that electroporation reduces tumor blood flow. In addition, we showed that electrochemotherapy, besides affecting tumor blood flow, act cytotoxic on tumor endothelial cells, contibuting to overall antitumor effectiveness of this therapy.

Based on the in vitro selection of siRNA molecules against oncogene k-Ras, we were the first to prepare plasmid DNA encoding miRNA against K-ras gene. In experimental tumor model of human colorectal carcinoma in mice we demonstrated silencing of the target gene and also effectiveness on tumor growth. As transfection method we used electroporation.

Electroporation and electrochemotherapy induce a profound reduction in tumor blood flow, which contributes to the antitumor effect. The antitumor effectiveness of electrochemotherapy has been proven extensively in the treatment of malignant melanoma, and now also exploited for palliative treatment of bleeding melanoma metastases. It demonstrated immediate cessation of bleeding and very good antitumor effectiveness which is of utmost importance for translation of electrochemotherapy into the treatment of deep-seated tumors, especially in well vascularized organs.

The effect of electrochemotherapy on metastatic potential of tumour cells is not known. In this study we demonstrated that electrochemotherapy with cisplatin does not affect migration and invasion of melanoma cells. Besides, electrochemotherapy with cisplatin does not affect melanoma cell adhesion to extracellular matrix proteins and also, it does not affect expression of genes, involved in cancer development. These results show that electrochemotherapy with cisplatin does not affect metastatic potential of malignant melanoma cells.
