(4R*,5R*)-4-benzoylamino-5-phenyl-3-pyrazolidinone : a useful building block in the synthesis of functionalized pyrazoles.

A review on 4-benzoylamino-5-phenyl-3-pyrazolidinone as a useful building block for the synthesis of functionalized pyrazoles containing an ?-amino acid or a dipeptide structural motif. The most useful derivatives are stable azomethine imines , obtained in the reactions with aromatic aldehydes. [3+2] cycloadditions of azomethine imines were studied. High selectivity of cycloadditions and simplicity of isolation of the products enable combinatorial solution-phase synthesis of pyrazolopyrazolone-type peptidomimetics with variable amino acid sequence and configuration.

Enantioselective preparation of [beta][sup]2-amino acid derivatives for [beta]-peptide synthesis.

ß-Amino acids with a single side chain in the ?-position (ß2-amino acids or H-ß2hXaa(PG)-OH; i.e., homo-amino acids with proteinogenic side chains) have turned out to be important components in ß-peptides. They contribute to unique secondary structures, they are required for mimicking the structure and the activity of ß-turn-forming ?-peptides, and they can be used for protecting ?-peptides against attack by aminopeptidases. In contrast to ß3-homo-amino acids, the ß2-isomers have to be prepared by enantioselective reactions or sequences or transformations, which are presented herein.
