We synthesized a new lanthanide single thiol reacting tag for the paramagnetic labelling of proteins, named Cys-Ph-TAHA, which gives NMR spectra with a single set of peaks. Bound to ubiquitin, it induced large residual dipolar couplings and pseudocontact shifts that could be measured easily and agreed very well with the protein structure. We show that Cys-Ph-TAHA can be used to label large proteins that are biochemically challenging such as the Lac repressor in a 90 kDa ternary complex with DNA and inducer.

Ammodytoxins are presynaptically neurotoxic secreted phospholipases A2 (sPLA2) from the long-nosed viper (Vipera a. ammodytes) venom. The exact molecular mechanism of action is still unknown, but it is clear that the toxicity is due to binding to the specific receptors in nerve endings of the motoneurons and its enzymatic activity. On the conference we presented 3D model of the complex between ammodytoxin and intracellular 14-3-3 proteins and calmodulin build based on the experimental results and explained possible rule of the 14-3-3 proteins and calmodulin as a stabilizators of enzymatic activity in reducing cytosolic environment.
