Slovenian Biochemical Society organized FEBS3 + meeting with participation of Biochemical Society of Croatia, Hungary and Serbia. Prof. N. Poklar Ulrih was president of the organization committee, prof . J. Kos was a Chairman of the Scientific Committee. The International Meeting was attended by 320 participants. During the congress four plenary lectures, 28 invited lectures, 5 corporate presentations, 39 short lectures and 159 posters were presented.

Predstavili smo rezultate raziskave stabilnosti antioksidantov v soku granatnega jabolka med pogoji simulirane in vitro prebave (želodec in tanko črevo). Antioksidativno učinkovitost (AOC) smo določali s luminescenčno metodo, skupne antociane (TA) s pH diferenčno metodo in skupne fenolne spojine (TP) s Folin–Ciocalteu-jevo metodo. AOC in TP se med in vitro prebavo nista znižala, ne glede na aktivnost pepsina. TA so ostali stabilni le med kislo želodčno fazo (pH 2), medtem ko se je njihova vsebnost med fazo tankega črevesa močno znižala (do 78%). Aktivnost pepsina je vplivala na TA v obeh fazah prebave, večje aktivnosti pepsina so vsebnost TA znižale bolj kot manjše. S pomočjo HPLC-DAD smo spremljali količino posameznih antocianov med simulacijo prebave ob prisotnosti in odsotnosti prebavnih encimov (pepsin, pankreatin). Prisotnost prebavnih encimov je le malo vplivala na vsebnost posameznih antocianov, največje zmanjšanje tako lahko pripišemo v glavnem spremembi pH, predvsem v fazi debelega črevesa, ki poteka pri pH 5,3.

COBISS.SI-ID: 4535160α-Synuclein is a small natively unfolded presynaptic protein which is implicated in the onset of Parkinson's disease and other neurodegenerative disorders. It is assumed that α-synuclein pathogenesis is associated with the process of fibrillation, while for its physiological function, its binding to synaptic vesicles seems to be of most importance. The present study investigates the mechanism of fibrillation and the lipid membrane binding of native recombinant α-synuclein and their mutants, Y39A and Y(125,133,136)A. We propose that the Tyr residues are important in particular for the early event of fibrillation, while their role in the protein-membrane interactions seems to be less significant. We confirmed the formation of α-helical structure upon binding of α-synuclein to the negatively charged small unilamellar vesicles in the liquid disordered phase. N-terminal part of the protein is at least partially inserted into the membrane. This protein did not associate with neutral lipids. These results indicate that in addition to electrostatic interaction, hydrophobic interactions are also important in the association of α-synuclein with membranes. The mechanism of α-synuclein binding to membrane is primarily dependent on the net charge of lipid heads, surface charge of the lipid bilayer, and the phase state of the lipids. We propose that α-synuclein has an ordering function and thermally stabilizes vesicles. We have confirmed the binding of α-synuclein with ganglioside GM1. We have also shown that the flavonoids quercetin, epigallocatechin gallate, and cyanidin-3-glucoside, inhibit α-synuclein fibrillation. Epigallocatechin gallate and nicotine are efficient in crossing the blood-brain barrier model and protect the neuron culture against oxidative damage.

The monograph was published in 1000 copies in two languages (Slovenian and Croatian).
