Abstract
Probiotics represent a possible alternative for the use of antibiotics as a feed supplement and in human nutrition in prevention or even in therapy of some diseases. Human bacterial isolates K7 and LF221 from Lactobacillus acidophilus group fulfil basic criteria for probiotic strains, produce bacteriocins with wide spectra of activity and are able to inhibit selected pathogenic bacteria. Study of the gut colonisation and of some functional effects (impact on faecal microflora composition, weight gain, general health state, nuclear DNA damage) will be performed on 24 weaned piglets. A combination of conventional microbiological and molecular-genetic methods (RAPD, PCR) will be applied for the tracing of a particular strain in mixed bacterial population of faeces. New isolates of lactic acid bacteria interesting for future use in species-specific probiotic preparations for animals will be obtained as well. Functional properties (adhesion, invasion) of both strains which are important from the point of view of safety and efficiency of human probiotics will be studied on human cell lines. The suitability of different media like cheese, milk or whey for the growth and survival of test probiotic bacteria and as a food vehicles will be tested. Their survival during drying ("spray-dry", "fluid-bed") will be examined as well. The results obtained will be useful for the development of probiotic preparations suitable for animal or human application.
