Significance for science
Detailed characterization of supports provided insight into adsorption-desorption processes of macromolecules, enabling development of predefined monolith microscopic structure and design of appropriate housing. Better understanding of adsorption phenomena also enabled prediction of specificity for certain chemical moieties as well as strength of interaction and consequently isolation yield. As an outcome, monolith bearing different chemical functionalities in predefined order on a single skeleton was prepared and characterized with proteins, representing novel approach in purification. Results obtained with test samples were further investigated with different important molecules like plasmid DNA and bacteriophages, later due to their geometrically defined structure and safety. Based on a pressure drop measurement data we were able to predict bacteriophage orientation after adsorption – another novel scientific outcome of this work.
Significance for the country
Program contributed for development of Slovenia in two fields: one is recognition of Slovenia as hi-tech country while the second is generation of new job positions. CIM monolithic supports are today already well established matrix in the field of chromatography recognized worldwide, as a commercially product but also as an scientific topic, always related to Slovenia. On the other hand, company BIA Separations, where this program was performed, generated in last 3 years over 20 new working positions, mainly for highly educated employees.
