Abstract
Yeast biomass production with high content of RNA by Saccharomyces cerevisiae ZIM-753 and Candida utilis ZIM-70 was lasting in aerobic batch conditions included selection and optimization of medium, and bioprocess parameters. According to results of screening methods of four C:N ratio (5:1; 10:1; 25:1; 50:1) and starting pH (2, 4, 6, 8) of media on the shaker the followed bioprocess parameters were selected for Saccharomyces cerevisiae: media with C:N ratio 5:1, starting pH 4.0, bioproces duration 15 hours, temperature 29°C, air flow 2 vvm and 250 rpm. In Candida utilis the media C:N ratio was 10:1, starting pH 6.0, and bioproces duration 12 hours. During the bioprocess in bioreactor the ''''on line'''' and ''''off line'''' measurements of pH, dry biomass, RNA, proteins, O2, CO2, glucose, nitrogen and organic acids were followed. In bioprocess of growth of Saccharomyces cerevisiae ZIM-753 the biomass concentration was 6.25 g/L and specific growth rate 0.24 h-1. The ratio of RNA and proteins in the yeast biomass was 7.3% and 39%, respectively. The concentraction of acetic acid and citric acid attained values 0.74 g/L and 0.1 g/L. The productivity for RNA production was obtained value 4.8 mg RNA/ g biomas h. In the case of Candida utilis ZIM-70 the concentration of biomass reached value 10.6 g/L at specific growth rate 0.4 h-1. RNA and proteins content in yeast biomass reached 4,9% and 34% while RNA productivity was in this case 4.1 mg of RNA/g biomas h.
