Significance for science
• The most important results of the research project L4 2042 are as follow:
SI P- 200,900,316 Patent enables the basement for new to technology procedure for increase the fermentative production of glycerol in wines. The process of introducing a new technology based introduction of thermal shocks on Saccharomyces cerevisiae inoculum cell suspension that reduce the number of inactive yeast cells, while others acquire memory effect (memory effect), allowing them to control the redox potentials during fermentation with the activation of metabolic pathways of glycerol production by affected yeast used in the fermentation process. This cells are hiper producing the glycerol as a oxireductive regulator of or the emergent situation. This procedure allows up to 80% increase in glycerol. The increased production of glycerol in wines gain fullness and commercially higher added value of the wines, allowing the importance of the market value of the final product. The process of increased production of glycerol can be applied in large scale.
• The procedure has been successfully tested in pilot and production on an industrial scale.
• In collaboration with the University of Auckland, New Zealand, in the determining of glutathione new method in our research based on HPLC-FD was successfully introduced and validated in practice. This method is now in regular practice in World's only five laboratories representing genuine achievement.
• Using SPME GC-MS, a new method for determination of volatile thiols (4MMP, 4MMPOH, 3MH, 3MHA) that give wine, fruity and tropical character was set up and validated and applied in our research
• Using SPME GC-MS, a new method for determination of metoxipirazines (IBMP, IPMP) and glutathione in samples of grapes set up and validated and applied our research.
• GC-MS were used in must and wine volatile compounds, higher alcohols and esters. Acquired knowledge and the results were included in agronomic profile of vineyards as we allow that we are on the basis of this knowledge to decide the best time for harvesting.
• Oxidation does not affect the metoxipirazine content. Influence of dissolved oxygen in the presence / absence of SO2 and volatile thiols, glutathione and metoxipirazine was studied. The use of sulfur dioxide was found as crucial in inactivating the oxidizing enzymes and consequently maintaining glutathione and volatile thiols. In the survey we conducted over 100 measurements of dissolved oxygen during the preparation and bottling of wine and introduce and examine ways of reducing by flushing with nitrogen.
• Analysis carried out technological processes has allowed selection of the most appropriate technology and analysis needed to maintain the varietal and aromaticity, we want to maintain throughout the course of all phases of grape must, fermentation of wine with distinctive varietal aromatic character. With this system we were able to create a characterization of selection of individual vineyard and make appropriate fermentativno this technology to maintain the original abilities of varietal aromatic varieties Sauvignon
